خلاصة اثنا عشر عزلة بکتیریة من عینات مختلفة من اللبن الرائب والحلیب تم شراؤها من الأسواق المحلیة فی محافظة الدیوانیة. تم إجراء الفحص الأولی لجمیع العزلات البکتیریة عن طریق صبغة غرام. تم اختیار ست عزلات بناءً على شدة التباین اللونی فی وسط التخمیر وتم التعرف علیها من خلال الفحوصات المظهریة والمیکروسکوبیة والکیمیائیة والحیویة والتی اشتملت على اثنی عشر عزلة من Lactobacillus spp. تم استخدام جهاز Vitek II Compact مع جمیع العزلات لتأکید التشخیص الأولی لجنس البکتیریا . تم إجراء الفحص الثانوی لعزلات من نوع Lactobacillus spp. بالنظر إلى کمیة حامض اللاکتیک المنتج ، کان لسلالة L. paracasei CAU 9836 أعلى إنتاج لحامض اللاکتیک ، والذی کان 64.32 ملجم / لتر. اتبع اختبار 16S rRNA لتشخیص جنس البکتیریا . أظهرت النتائج أن أفضل الظروف لإنتاج الحامض کانت 62.76 ملجم / لتر بواسطة L.paracasie 9836 المنماة على وسط بدیل یحتوی على 50٪ شرش ، و 7٪ دبس السکر(المولاس) ، ودرجة الحموضة 6.9 ، وفی حاضنة هزازة بسرعة 250 دورة فی الدقیقة عند 38 درجة مئویة لمدة 24 ساعة . تم استخدام تقنیات FTIR للتعرف على نواة ذرة الهیدروجین (البروتون) 1H وذرات الکربون 13C فی وجود حامض اللاکتیک القیاسی للمقارنة. أظهرت النتائج أن قمم الحامض المنتج کانت متشابهة إلى حد ما مع تلک الموجودة فی الحامض القیاسی .
